New monoclonal antibody developed that can target proteins inside cancer cells

Mar. 13, 2013 ? Researchers have discovered a unique monoclonal antibody that can effectively reach inside a cancer cell, a key goal for these important anticancer agents, since most proteins that cause cancer or are associated with cancer are buried inside cancer cells. Scientists from Memorial Sloan-Kettering Cancer Center and Eureka Therapeutics have collaborated to create the new human monoclonal antibody, which targets a protein associated with many types of cancer and is of great interest to cancer researchers.

Unlike other human therapeutic monoclonal antibodies, which can target only proteins that remain on the outside of cancer cells, the new monoclonal antibody, called ESK1, targets a protein that resides on the inside of the cell.

ESK1 is directed at a protein called WT1, which is overexpressed in a range of leukemias and other cancers including myeloma and breast, ovarian, and colorectal cancers. WT1 is a high priority target for cancer drugs because it is an oncogenic protein, meaning that it supports the formation of cancer. In addition, it is found in few healthy cells, so there are less likely to be side effects from drugs that target it.

"This is a new approach for attacking WT1, an important cancer target, with an antibody therapy. This is something that was previously not possible," said David A. Scheinberg, MD, PhD, Chair of the Sloan-Kettering Institute's Molecular Pharmacology and Chemistry Program and an inventor of the antibody. "There has not been a way to make small molecule drugs that can inhibit WT1 function. Our research shows that you can use a monoclonal antibody to recognize a cancer-associated protein inside a cell, and it will destroy the cell."

The first studies of the antibody are showing promise in preclinical research as a treatment for leukemia as reported March 13, 2013, in Science Translational Medicine.

"ESK1 represents a paradigm change for the field of human monoclonal antibody therapeutics," said Cheng Liu, PhD, President and Chief Executive Officer of Eureka Therapeutics. "This research suggests that human antibody therapy is no longer limited to targeting proteins present outside cancer cells, but can now target proteins within the cancer cell itself."

ESK1 was engineered to mimic the functions of a T cell receptor, a key component of the immune system. T cells have a receptor system that is designed to recognize proteins that are inside the cell. As proteins inside the cell get broken down as part of regular cellular processes, molecules known as HLA molecules carry fragments of those proteins -- known as peptides -- to the surface. When T cells recognize certain peptides as abnormal, the T cell kills the diseased cell.

In the current study, the investigators showed that ESK1 alone was able to recognize WT1 peptides and kill cancer cells in the test tube and also in mouse models for two different types of human leukemia. "We were surprised that the antibody worked so well on its own," said Dr. Scheinberg, senior author of the paper. "We had originally expected that we might need to use the antibody as a carrier to deliver a drug or a radioactive therapy to kill the cancer cells, but this was not necessary."

Additional studies must be done in the laboratory before ESK1 is ready to be tested in patients. But the monoclonal antibody was engineered to be fully human, which should speed the time it takes to move the drug into the clinic. Researchers expect that the first clinical trials, for leukemia, could begin in about a year.

The antibody was developed under a collaborative effort between Memorial Sloan-Kettering and Eureka, which have jointly filed for patent protection.

Nigerian extremists claim they killed 7 hostages

KADUNA, Nigeria (AP) ? A breakaway Islamic extremist group said Saturday it had killed seven foreigners who its members kidnapped from northern Nigeria, according to an online message purportedly from the group.

The message, identified as coming from Ansaru, could not be immediately verified by The Associated Press, though it included photographs the group claimed showed the dead, who were kidnapped from a construction company compound in February. Those kidnapped included four Lebanese citizens and one each from Britain, Greece and Italy ? all employees of Setraco, a Lebanese construction company with an operation in Bauchi state, officials said.

Two Nigerian military spokesmen declined to immediately comment when reached by the AP, while a presidential spokesman and a spokeswoman for the country's domestic spy service could not be immediately reached. In a statement later Saturday, the British Foreign Office said it was "urgently investigating" the killings claimed by the group.

The message, posted to an Islamic extremist website Saturday, said Ansaru members killed the hostages after British warplanes were reported to have been seen in the northern Nigerian city of Bauchi by local journalists. However, the reports referenced in the online statement actually referred to the airplanes being seen at the international airport in Abuja, Nigeria's capital.

"As a result of this operation, the seven hostages were killed," the group said in the statement. It said a video of the killings would be posted online. An online image accompanying the posting appeared to show a gunman standing over dead bodies.

Ansaru said its decision to kill the hostages was also sparked by a statement immediately after the abduction from Nigerian President Goodluck Jonathan who said the government would do anything in its power to free the hostages. The group said security forces had recently arrested an unspecified number of its members.

Ansaru previously issued a short statement in which it said its fighters kidnapped the foreigners Feb. 16 from a construction company's camp at Jama'are, a town about 200 kilometers (125 miles) north of Bauchi, the capital of Bauchi state. The attack saw gunmen first assault a local prison and burn police trucks, authorities said. Then the attackers blew up a back fence at the construction company's compound and took over, killing a guard in the process, witnesses and police said.

The gunmen appeared to be organized and knew who they wanted to target, leaving the Nigerian household staff at the residence unharmed, while quickly abducting the foreigners, a witness said. Local officials in Nigeria initially identified one of the hostages as a Filipino, something the Philippines government later denied.

In January 2013, Ansaru declared itself a splinter group independent from Boko Haram, the north's main Islamic terrorist group, analysts say. Boko Haram, whose name means "Western education is sacrilege," has launched a guerrilla campaign of bombings and shootings across Nigeria's predominantly Muslim north. Boko Haram is blamed for at least 792 killings last year alone, according to an AP count.

On Saturday, a military spokesman said at least two soldiers and 52 Boko Haram fighters were killed in Maiduguri in fighting after a visit by the president. Those tallies could not be independently confirmed by the AP. Security forces often downplay their casualties, as well as those of civilians in the ongoing fight with guerrillas.

Britain has previously linked Ansaru to the May 2011 kidnapping of Christopher McManus, who was abducted with Italian Franco Lamolinara from a home in Kebbi state. The men were held for months, before their captors killed them in March 2012 during a failed Nigerian military raid backed up by British special forces in Sokoto, the main city in Nigeria's northwest. That rescue mission strained Italian-British relations, and out of it came pledges from Italy and Britain to beef up their cooperation on security matters and share information.

Ansaru earlier claimed the kidnapping in December of a French national working on a renewable energy project in Nigeria's northern Katsina state. Meanwhile, a group of men claiming to belong to Boko Haram claimed responsibility for the kidnapping of seven French tourists from northern Cameroon late February ? a first for the group.

Analysts say Ansaru likely has closer ties to al-Qaida in the Islamic Maghreb, the Africa branch of the terror network. Analysts have warned that there likely will be more attacks by Ansaru targeting Westerners and Western interests in Nigeria, as well as neighboring nations. Ansaru initially said it carried out the kidnappings in part as a response to the French military intervention in Mali. The French, backed up by Malian soldiers, have been fighting the Islamic militants who seized control of northern cities there in the weeks following a coup that toppled the West African nation's democratically elected government.

Despite the deployment of more soldiers and police to northern Nigeria, the nation's weak central government has been unable to stop the killings. Meanwhile, human rights groups and local citizens blame both Boko Haram and security forces for committing violent atrocities against the local civilian population, fueling rage in the region

Sony Xperia Z was announced back at CES in January and its popularity grows bigger by the day. We have already had the Sony Xperia Z for quite a while in our office and its in-depth review is over here but once the MWC wrapped up we were also able to complete its battery test.

The smartphone has a large 2330 mAh battery to rely on, but the quad-core Krait CPU and the 5? 1080p display are known to consume quite a lot of power. The fact that the battery isn?t user-replaceable puts even more pressure on the Xperia Z to do well here, and we were as curious as you are to find out if that?s actually the case.

Our battery stress tests begin with 3G talk time. Sony claims the Xperia Z will last 14 hours of 3G calls before a fully charged battery goes flat. However, the smartphone managed to pull off a surprisingly great performance here and topped this score easily.

The Sony Xperia Z we had in our office managed to last 16 hours and 3 minutes on 3G talks. We tested two different units and they both did similarly here, so it?s not a lucky exception, either. It?s quite rare for a phone to outdo its official rating, and the Sony Xperia Z couldn?t possibly hope for a better start here.

Unfortunately, the Sony Xperia Z wasn?t quite as impressive when it came to video playback endurance. With the 5? 1080p LCD coming into play the battery lost 90% of its charge in just 5 hours and 39 minutes. It?s still enough to watch two full movies in one go, but it?s one of the worst performances we have seen here nonetheless. We ran the test with Mobile BRAVIA 2 turned on as we believed that?s how most users will be using their smartphones, but even when we switched it off we didn?t notice any big difference in the Xperia Z battery life.

The Sony Xperia Z managed to climb up the ranks in the web browser test. It lasted for almost an hour longer compared to the video playback chapter as, unlike the AMOLED screens out there, its LCD screen consumes no extra power to display the predominantly white web pages. 6 hours and 37 minutes still isn?t something to write home about, but it?s still a decent achievement and better than the Oppo Find 5 and HTC Butterfly, which are the other two 1080p 5-inchers we have tested.

As you would expect from with an ample battery like that, the stand-by performance of the Sony Xperia Z is quite good and helped it to a pretty good final endurance rating of 48h. Unsurprisingly, the Sony Xperia Z can last ages when the screen is not active (in-call and stand-by), but the display is quite power-hungry. Still, according to our result you will only need to charge your smartphone every 48 hours if you do an hour of each of the three tested activities each day, which is quite good.

News Corp gives publishing company $2.6 billion in cash

(Reuters) - Rupert Murdoch's News Corp will start its largely print-based publishing company with no debt and $2.6 billion (1.7 billion pounds) in cash when it completes the spinoff.

The amount of cash and debt, which analysts generally expected, gives the publishing company a purse to buy other assets should it choose. News Corp released the details in a regulatory filing on Friday.

"You can never count out Murdoch when it comes to buying companies," said Gabelli & Co analyst Brett Harriss.

News Corp said in the filing the publishing company's assets are worth $18.6 billion (12.46 billion pounds), which includes newspapers such as The Wall Street Journal and Times of London, book publisher HarperCollins and its ownership stake in other assets such as Australian pay-TV company Foxtel.

Media businesses, especially anything related to print, are going through major ownership changes. Tribune Co is exploring the sale of its newspaper assets, which include the Los Angeles Times and Chicago Tribune. Time Warner Inc is spinning off Time Inc, home to magazines Sports Illustrated, Fortune, People and its eponymous newsweekly.

Analysts estimate that Time Inc, which is expected to become a stand-alone public company at the end of the year, will be worth about $2 billion (1 billion pounds) to $3 billion (2 billion pounds).

News Corp's publishing operations "certainly have the fire power to buy Time Inc," Harriss said.

News Corp declined to comment on acquisition opportunities.

PHONE HACKING TAB

News Corp is injecting money into the new publishing company as the print industry faces unrelenting declines in readership and advertising revenue.

The $2.6 billion (1.7 billion pounds) in cash is more than three times the $741 million (496 million pounds) that the publishing division had on hand as of December 31.

At that time, News Corp as a whole had $7.8 billion (5.2 billion pounds) in cash and cash equivalents and a little more than $16 billion (10.7 billion pounds) in short- and long-term debt.

The publishing division had revenue of $4.16 billion (2.78 billion pounds) for the six months ending December and an operating profit of $291 million (195 million pounds).

Last June, the global media conglomerate announced the plan to split its publishing and entertainment assets into two publicly traded companies.

The publishing company will retain the News Corp name. The entertainment businesses, which include the 20th Century Fox film studio, Fox broadcasting network and Fox News channel, will be called the Fox Group.

The spinoff company will be traded on the Nasdaq, while the shares will also be listed on the Australian Securities Exchange. The trading symbol is still unknown.

News Corp also said the Fox Group would be responsible for any payments in the civil cases and any investigations involving the telephone hacking scandal at its British newspapers.

For the past several years, News Corp has been dealing with the fallout from the scandal, which stemmed in large part from its now-defunct News of the World.

On Friday, a former policeman and an ex-prison officer admitted to selling information to the Murdoch-owned tabloid the Sun.

News Corp shares were up 2.1 percent at $30.59 in afternoon trading on the Nasdaq.

Growing Home: DIY: Glitter-Block Easter Eggs

If you've been reading this blog for awhile, you know well that I can't resist layering things in gold. Gold leaf, gold paint, gold home accents...I'm even totally exclusive about gold jewelry (silver kind of washes me out). So when the egg dyeing bug hit me last week, gold glitter came instantly to mind. I found that blocking the glitter against bright spring colors gives the eggs a modern, geometric look without sacrificing any of the drama of the gold. Which means that now we have a collection of Easter eggs that ?is equal parts bright, happy, and blinged out:). Even the boys like to ooh and aahh at them, which of course makes me love them even more.?

These eggs take very little time and can be made from things laying around your house (that is, if you keep glitter on hand). Below, I've given you the basic process as a photo tutorial (yup, that's another something new!), but I did not tell you how to go about making egg dye. The short story is that it's easy: combine roughly 1/2 cup hot water with a teaspoon of white vinegar and 20 drops of food coloring. For the long story, consult Martha?here. You'll notice that I've also blown out the eggs so that they can stay out of the fridge as holiday decoration and be kept from year to year. The blowing process is awkward the very first time you do it, but quite easy once you get a system down (I only broke one). The key is to do it after the dye bath, as a hollow egg won't submerge!

To make your own glitter-block Easter eggs, follow the steps below. I think you'll find creating them to be fairly addicting.

Materials: raw eggs, embroidery needle (or large sewing needle), egg dye, school glue, paintbrush, metallic glitter and an extra bowl

Now, use your lips or a rubber bulb syringe (like the ones they give you at the hospital after you have a baby) to blow into the small hole, emptying the egg into your extra bowl. The bigger you've made the large hole, the easier it will be. The photo above shows you about the right size.?

Couple allegedly steals 2 cop cars during chase

The young couple accused of stealing multiple police cars from two cities and leading authorities on a high-speed chase through two states Tuesday morning have been identified as Blake Bills and Shayna Sykes.

"I never heard of anyone stealing two police cars in one incident," said Philadelphia Deputy Police Commissioner Richard Ross.

Camden, N.J., Police Chief John S. Thomson tells NBC10's Cydney Long?that Camden Police Officer?Sekou Reid-Bey?made a traffic stop at 9:49 a.m., directly in front of Camden Police Headquarters at 800 Federal Street, when the incident began.

"As [the officer] was interviewing the driver of that car, he heard his car door shut," he said.

Thompson says Reid-Bey, 49,?saw the couple jump into his cruiser. He says they struck him, breaking the officer's leg,?before driving off.

Initially, Sykes, 23,?and Bills,?24, were pursued by both the Camden and Pennsauken Police Departments -- reaching 100 mph on the Admiral Wilson Boulevard, according to Camden County prosecutors. That pursuit lasted?about 10 minutes?and wove through the city of Camden and portions of Pennsauken.

As the stolen police cruiser crossed the Ben Franklin Bridge the South Jersey officers peeled off and Philly PD?picked up the?chase.?The suspect eventually crashed the Camden Police car in the area of 7th and Norris in North Philadelphia.

Sykes fled from the crashed cruiser, according to law enforcement sources, leaving Bills trapped inside the Camden Police car.

With police attempting to apprehend Sykes, sources say she managed to steal a Philadelphia Police car, No. 2625, from the area of 7th and Norris, also in Philadelphia.

As it was being chased, the stolen Philadelphia cruiser hit three cars at 6th and Lehigh. It also?nearly struck?a pedestrian walking near 5th and Clearfield. That person fell and hit their head -- his/her?condition is unknown.

The car was eventually stopped at 1100 Hope Street on the edge of Northern Liberties. Sykes attempted to run from the cruiser, but was apprehended by several officers just feet from the stolen car.

Sykes was handcuffed and put into another police cruiser. She was taken to East Detectives. The entire Philadelphia chase lasted about 15 minutes.

"I thought it was a movie," Felix Vargus, a tow truck driver, told NBC10's Claudia Rivero. "She was trying to fight with them so she could escape again."

Another witness who observed her arrest told Rivero the woman was not wearing shoes when she was taken into custody.

Police say that drugs possibly played a role in the incident and that both Bills and Sykes will face a slew of charges including resisting arrest, DUI, fleeing police and risking a catastrophe.

"And that's just here," Thompson said. "The other charges you will need to deal with Camden prosecutors for that."

Sykes and Bills are suspected of evading an officer during an attempted traffic stop Sunday -- just two days before the high-speed chase.

Camden prosecutors say they expect to charge the duo with aggravated assault and theft.

Reid-Bey was being treated at Cooper University Hospital.?

"He's doing well. He's going to be fine. He's in stable condition; significant injuries to the lower extremities of his body though," said Thompson. He says the officer, after being hit, went onto the cruiser's hood before being thrown to the concrete. The injured officer was then able to radio for assistance.

An official with the Camden Fraternal Order of Police tells NBC10 that Reid-Bey is an 18-year-veteran of the force.

"There will be a litany of charges she will face in Philadelphia," said Ross. "I think [authorities] did a great job under the circumstances."

Bills and Sykes both hail from Macungie, Pa. Police say that Bills grandmother reported the couple missing earlier in the day after they allegedly took the woman's car and left her with the couple's 7-month-old child.?Police also suspect the couple is involved in another police chase on Sunday that left an officer injured.?

Lawyers for Gitmo prisoners decry 'alarming' conditions

Lawyers for terror suspects held at the U.S. detention facility at Guantanamo, Cuba, said Tuesday that detainees are engaged in widespread protests of conditions at the prison, including a hunger strike that may imperil their lives.

Calling the situation ?alarming,? the lawyers said in a statement that some of their clients are ?coughing up blood? and ?losing consciousness.?? A letter making similar assertions was sent earlier this week to Navy Rear Adm. John W. Smith, the commander of the Joint Task Force at Guantanamo and signed by a dozen lawyers who represent most of the detainees at Guantanamo.??

A spokesman for U.S. military at Guantanamo?? disputed the lawyers? claims of a widespread hunger strike, saying they and their clients were merely trying to get attention and keep Guantanamo ?in the news. ??

The spokesman, Navy Capt. Robert Durand, said that a half-dozen detainees are currently on a hunger strike -- five of whom are being force fed through tubes -- and that no lives were in danger. Durand added that the figure was consistent with the average number of hunger strikers at Guantanamo over the past several years. He also acknowledged that ?some detainees? have been disciplined and moved out of Camp 6 -- the most permissive of the camps at Guantanamo, with communal living arrangements -- but he declined to say how many or give the reasons for the action.?


The conflicting claims underscored the difficulty of obtaining information about conditions at the facility, which President Barack Obama vowed to shut down on his first day in office after his 2008 election but which still remains open as a result of congressional opposition to its closure. There are 166 detainees remaining at the camp, but military rules forbid them from communicating in any way with members of the news media and visits to the camp by outsiders are tightly regulated. Even their communications with their lawyers must be cleared by military censors.

One of those lawyers, David Remes, told NBC News in a telephone interview from Guantanamo Monday night that he saw one of his clients -- Hussain Almerfedi, a Yemeni -- earlier that day and that he had lost ?substantial weight? and was ?very sick.? Under Guantanamo rules, Remes said he could not share anything that his client told him until the censor cleared the communication. One flashpoint appears to have been a Feb. 6 search at Camp 6 in which, according to the lawyers, camp authorities seized blankets, sheets, towels, sleeping mats, razors and other items from the detainees,? including family photos and religious CDs from the detainees. In their letter to Smith, the lawyers alleged that Arabic interpreters at the camp inspected Qurans ?in ways that constitute desecration.??

Durand, the Guantanamo spokesman, disputed that any harsher restrictions had been imposed by the new commander and said the search last month was in keeping with past practice. He said that search, and earlier ones, have turned up? ?a Wal-Mart worth of stuff,? including improvised weapons, illegal electronics and other illicit contraband. But he said that handling of the Qurans was tightly regulated? and that no guards are even permitted to touch the Islamic Holy Books during the searches.

Durand also acknowledged that some of the dispute between camp authorities and the detainees? lawyers may be about defining terms. Guantanamo officials define a hunger strike as refusing to eat nine meals in a row. But, he said, some of the detainees may be hoarding food in their cells even when they claim to be on a hunger strike.??

Gate placement a concern for NASCAR after crash

(AP) ? NASCAR will look at the placement of gates at its tracks after a Nationwide Series car crashed through the fence at Daytona and injured more than two dozen fans.

The fans were injured during the 12-car crash last Saturday when pieces of rookie Kyle Larson's car ripped through the fence, including a section where a gate connects the grandstand and the track.

"I think because of where it came through and having pieces that did get through and it being a gate area, that's really going to be the focus for us to look at," Steve O'Donnell, NASCAR's Senior Vice President, Racing Operations said Saturday from Phoenix International Raceway. "We're certainly going to look at fencing in general, but I think that particular area, that it was a gate, did impact it. We know the gate was locked, but does that provide as much stability as the rest of the fencing we believed it did? We've now got to look at that impact."

The crash occurred on the final lap of the Nationwide race the day before the Daytona 500, when leader Regan Smith tried to block Brad Keselowski and triggered a chain reaction. Larson's car went airborne during the wreck and slammed into the fence, sending debris into the stands.

O'Donnell said two injured fans remain at the hospital, but everyone else has been released.

The crash has forced NASCAR to take a closer look at its safety measures, particularly fencing around its tracks.

O'Donnell said NASCAR mostly leaves fencing up to the individual tracks, but may look at being more involved, similar to what it did requiring impact-absorbing SAFER barriers along concrete walls.

"It's important to note that most of the safety elements in that car did their job," O'Donnell said. "The driver, as you saw, walked away. However, the car then got up into the fence. Our focus is going to be if the elements in the car did their job, now what do we need to do in looking at the impact of the fence, what happened when that car impacted the fence with parts getting away."

O'Donnell also said that the tethers that hold Larson's car together worked, but that the section the tethers were attached to sheared the car, sending pieces flying.

"We've tethered a number of different things as we've learned and added safety aspects to the car, but what do we need to do in addition to that when we look at this aspect specifically?" O'Donnell said. "

Instead of bringing Larson's car immediately back to NASCAR's research and development center in Charlotte, N.C., it was left in Daytona so track officials use it in their investigation into what happened with the fencing. The car is in the process of being brought back to the R&D center, where it will be put back together and, with the help of video, hopefully determine what parts of the car came off when.

O'Donnell said NASCAR will bring in Larson's race team, which hasn't seen the car since it was impounded after the wreck, to talk about how the car was constructed and fabricated.

NASCAR also will bring in outside experts, including Dr. Dean Sicking, director of the Midwest Roadside Safety Facility at the University of Nebraska, officials from Indianapolis Motor Speedway and outside engineers to look at the fencing.

Daytona will have its experts work with an outside firm to analyze what was in place and analyze what may need to be done.

"When you talk about safety, I think Jeff Burton said it best: There's no end goal of safety, it's something we work on each and every day," O'Donnell said. "Same with this process. If there's something we can learn today, we'll apply that. If it's two months from now, we'll apply it as well.

O'Donnell said NASCAR will look at the issue of restrictor-plate racing at Daytona and Talladega, where the series stops in May, but he said he's comfortable with plate racing at the two superspeedways right now.
